Identifikasi Molekuler Isolat Bakteri Endofit dari Tanaman Porang (Amorphophallus muelleri Bl.) Sehat Berdasarkan Analisis Gen 16S rRNA

Penyakit busuk umbi yang disebabkan oleh Dickeya chrysanthemi menjadi salah satu ancaman utama terhadap kesehatan tanaman porang (Amorphophallus muelleri Bl.), sehingga diperlukan upaya pengendalian berbasis agens hayati. Bacillus aerius berpotensi sebagai bakteri pemacu kesehatan tanaman dan antagonis patogen, namun identifikasi spesiesnya memerlukan pendekatan molekuler karena kemiripan karakter fenotipik antarspesies dalam genus Bacillus. Tujuan penelitian untuk mengidentifikasi secara molekuler isolat bakteri yang dieksplorasi dari tanaman porang sehat yang tumbuh di antara populasi tanaman yang diduga terinfeksi busuk umbi akibat Dickeya chrysanthemi melalui analisis gen 16S rRNA, dan menentukan tingkat kemiripan sekuensnya dengan data pada basis data GenBank sebagai dasar penetapan identitas taksonomi isolat. Penelitian dilakukan menggunakan metode identifikasi molekuler melalui analisis gen 16S rRNA yang meliputi isolasi DNA, amplifikasi PCR, sekuensing, dan pencocokan sekuens terhadap basis data GenBank untuk menentukan identitas taksonomi isolat bakteri. Hasil penelitian menunjukkan bahwa isolat bakteri endofit dari tanaman porang sehat menunjukkan kemiripan genetik tertinggi dengan sekuens yang terdaftar sebagai Bacillus aerius pada basis data GenBank. Keberadaan bakteri pada tanaman porang sehat di antara populasi tanaman yang diduga terinfeksi busuk umbi akibat Dickeya chrysanthemi mengindikasikan bahwa bakteri ini memiliki mekanisme perlindungan alami yang berpotensi ekologis dalam mendukung kesehatan tanaman dan berpeluang sebagai agens pengendali hayati berbasis mikroba endofit.
